Objective The goal of this project is to design 20-µL cell-free protein synthesis (CFPS) reactions for sfGFP, PETase/TfCut2, and Reteplase that reduce reagent cost while maintaining or improving production of functional protein. The three targets provide an increasing challenge in folding: sfGFP is a robust reporter with no disulfide bond, TfCut2 contains one disulfide bond, and Reteplase contains nine disulfide bonds and therefore requires substantially stronger control of the oxidative folding environment.
