Day to day that means isolating nuclei from mouse tissue, immunostaining histone marks such as H3K27me3 and H3K27ac, and running high-content imaging in 384-well plates, with the image analysis on an HPC cluster.

Week 1 HW: Designed Mini-Protein Epigenetic Modifiers

Sep 22, 2026 · @Rishi Shetty
1. What I would like to biomanufacture, and why? The product. I propose to biomanufacture a library of designed mini-protein binders — engineered proteins of roughly 60–120 amino acids — that disrupt PRC2 by wedging into the interface between its EZH2 and EED subunits. PRC2 is the complex that deposits and propagates H3K27me3: EZH2 is the catalytic writer, and EED is the reader that binds existing H3K27me3 and allosterically stimulates EZH2 to write more, so the mark drives its own spread along chromatin. Breaking the EZH2–EED association collapses that read–write loop without abolishing basal methyltransferase activity.

Final Project
