What you would like to biomanufacture and why? Target Product: A commercial-grade, purified cell-free plasmid DNA amplification toolkit based on the ColE1 origin of replication (a “ColE1-Ciro” kit).
Motivation: The goal is utilitarian: to create a GMP-compliant, highly scalable alternative to bacterial fermentation for manufacturing therapeutic DNA. Current plasmid production for mRNA vaccines and viral vectors requires massive bioreactors and extensive downstream purification to remove toxic bacterial endotoxins (LPS) and genomic DNA. By shifting from a crude lysate to a completely defined mixture of purified proteins, we can manufacture endotoxin-free, pharmaceutical-grade circular DNA in a benchtop tube, drastically lowering the barrier to entry for gene therapy manufacturing.
